This kit is a steptococcus grouping test using nitrous acid extraction for the rapid identification of B-haemolytic streptococci of Lancefield types A, B, C, F and G.

Instructions

Extract
1a. Add three drops of Extraction Reagent 1 to a test tube. Add three drops of Extraction Reagent 2 to the same test tube. The resulting color is a light orange indicating that the nitrous acid solution is ready
1b. Pick 2-5 selected colonies and emulsify thoroughly in the extraction solution. There is no incubation step.
1c. Add three drops of Extraction Reagent 3 to the test tube. The resulting color is a light blue indicating that the extraction procedure is complete.

React
2a. Add one drop of extract to each circle and using a separate mixing stick for each, mix contents thoroughly, taking care to cover the entire area within the circle.
2b. Gently rock the card for one minute and observe for agglutination.
